Premier League leaders Floriana and Valletta were handed seemingly comfortable ties when they were drawn against Division One teams Mellieha and St Patrick respectively.
Six top-flight clubs and the ten Division One teams were in the hat for yesterday's draw at the Centenary Hall.
Last year's winners Hibernians, Marsaxlokk, Sliema Wanderers and Birkirkara were awarded a bye and enter the competition in the quarter-final stage which will be played in April.
Floriana, winners of the competition 18 times, should find little problems to book their place in the second round when they meet Mellieha who were promoted to Division One last season.
Valletta, who have won the FA Trophy 11 times, should prove too strong for St Patrick.
A more tricky tie was handed to Hamrun Spartans who were drawn to face Mosta. The Blues are targetting promotion to the Premier League this season.
The other Premier League teams in the draw were Pietà Hotspurs, who face St George's, Mqabba, who take on Vittoriosa, and Msida St Joseph, who play Senglea Athletic.
The ties will be played on November 3, 4, 10 and 11 at the Centenary Stadium.
