The Belgian, ranked fifth before the Roland Garros tournament, beat Russia's Svetlana Kuznetsova 6-4 6-4 to win her third title on Paris clay.
Henin-Hardenne was world number one in 2003 before injuries and a virus disrupted her career. She retired from the Australian Open final in January against Amelie Mauresmo because of illness.
France's Mauresmo remains at the top of the rankings ahead of Belgian Kim Clijsters.
Kuznetsova climbs to sixth from 10th. Czech teenager Nicole Vaidisova, knocked out in the semi-finals by Kuznetsova, rises three places to 13th.
In the men's rankings, Switzerland's Roger Federer remains number one ahead of Rafael Nadal. David Nalbandian of Argentina is third in an unchanged top five following Roland Garros.
